I tried to drag my entire applications folder to a new back-up drive but
when placing it in the window got what looked like a puff of smoke on
the screen and now the folder does not show though it is still there.
How do you get it to show up again?

If the Application folder was located in the left hand sidebar, then you
were dragging an alias of your Application folder. You can drag any
folder to the side bar and when you drag it off to get rid of it it goes
up in a puff of smoke.

Was the "folder" located in the Dock?

Probably in the side bar.

No ordinary alias. A good ordinary honest alias has some substance, like
an empty folder in a filing cabinet. You can move it about. You can lose
it and find it. You can roll it up and pretend to swallow it to amuse
the kids. You can also sometimes go wrong by burning an alias that
points to a substantial folder to a CD or whatever and finding it empty.

But a sidebar icon or folder or a dock one is altogether more
insubstantial, it is a nervous ghost of a thing and prefers suicide to
relocation. Which reminds me:

Two elderly spinster sisters were sitting in their
rocking chairs on their front veranda when
suddenly they saw a hen being chased by a cockerel
on the other side of the country road. The hen
bolted to the road but a truck ran it over. One
sister turned to the other with a knowing look and
said, "See! She would rather die!"
